چکیده

15 A comprehensive comparative analysis of the structure – antifungal activity relationships 16 for the series of biosynthetically engineered nystatin analogues, their novel semisynthetic 17 derivatives, as well as amphotericin B (AMB) and its semisynthetic derivatives was performed. 18 The data obtained revealed the significant influence of the structure of the C7 – C10 polyol 19 region on the antifungal activity of these polyene antibiotics. Comparison of positions of 20 hydroxyl groups in the antibiotics and in vitro antifungal activity data showed that the most 21 active are the compounds in which hydroxyl groups are in the positions C8 and C9 or C7 and 22 Copyright © 2013, American Society for Microbiology. Hydrazides--a novel type of derivatives of polyene macrolide antifungal antibiotics.

properties ofpolyene macrolides wehave established the structure-activity relationship among the derivatives of amphotericin B, the polyene antibiotic most commonly used in clinical practice11}. It has been shown in these studies that the improvement of selective toxicity is related to the absence of a free carboxyl group in the modified antibiotic molecule. Wehave also demonstrated that water ...

Molecular modelling of membrane activity of amphotericin B, a polyene macrolide antifungal antibiotic.

Amphotericin B (AmB) is a well known polyene macrolide antibiotic used to treat systemic fungal infections. Despite its toxicity AmB is still regarded as a life-saving drug. The lack of adequate knowledge of the AmB mechanism of action is a serious obstacle to efficient development of new less toxic derivatives. Complementary to various experimental approaches, computational chemistry methods w...

Steroid interference with antifungal activity of polyene antibiotics.

Wide differences exist among the polyene antibiotics, nystatin, rimocidin, filipin, pimaricin, and amphotericin B, with reference to steroid interference with their antifungal activities against Candida albicans. Of the numerous steroids tested, ergosterol was the only one which effectively antagonized the antifungal activity of all five polyene antibiotics. The antifungal activities of nystati...

Effect of fatty acids on action of polyene antibiotics.

Fatty acids cause a decrease in the absorption spectra of the antifungal polyene macrolide antibiotics nystatin, filipin, candicidin, and amphotericin B. For nystatin, filipin, and candicidin, this decrease in absorption could be correlated with the activity of the fatty acids in protecting the yeast Saccharomyces cerevisiae against the action of these antibiotics. With amphotericin B a correla...
